OBJECTIVES AND ACTMTIES
Obje¢tive$ and alms
History
The Foundation was firsl established in the USA in 2010 by three siblings who kjsl their mother, aged 53, to
stage IV HPV-related anal cancer. Frustrated by the limtted Iherapeutic and community support available to
her, they created the organisation to fill the void caused by s(2nt public, suentrfic and medical focLJS on this
disease and its cause, the human papillomawrus IHPVI. HPV causes anal cancer and at least five other
¢an¢ers, amounting to 50/¢ of ¢an¢ers woddwide. These cancers a￿ preventable. In 2012, the org8ni2ation
expanded to the UK with 2013 as its first full operational year.
Mission
The HPV and Anal Can¢er Foundati￿ empowers anal ¢an¢er pab.ents and a¢¢elerates prevention and
research methods that eliminate anal can￿r and the wnJs that causes the majority of cases, HPV.
Objectives
Highlight the relevance of human papillomavirus IHPVI in contracting can￿r,.
Advocate for prevention melh(Kls that wll end anal cancer and HPV-related cancers, including
gender-neutral vaccination-
Accelerate a cure for the 5% of all Can￿rS that are caused by HPV, wth a f￿uS on anal cancer. and
Providè support for sutwvors, caregivèrs, suèntsts and providèrs connÈctéd to anal can¢Èr.
The Need
A fundamental objective of the Foundation's ￿￿)r￿ is to reduce the inudence of anal cancer and other
HPV-related cancers. HPV is a very common skin wms that nearty every sexLJalty active person will get at
some point in their lives. HPV affects both men and women and is resp(￿sible for 5 /0 of the world's cancer
burden. HPV cau$8S fAnoers of the anus. ￿[￿x. head and neck. ￿nIS. vagina. and vulva.
Protection against HPV through unwersal HPV vaccinatK)n is a cribcal public heaAh goal. With HPV identified
as a cause of six different types of cancer, and 5% of cxncer Gases worldwde, li is imperative that we protect
the entire population against it. The most effective method to reduce Can￿rS related to HPV is to prevent
in((ial infection of the virus through Immunisab"on. The Foundab"on achieves this important airll through its
efforts to advance eqLJal access to HPV protection for tK>th boys and gitls through gender-neutral vaccination
against the virus. At the Intr￿UCtion of the vaccine in most countries, onty girls were able lo receive this
lrfe-sawng intervention Whi￿ boys were generally excluded. The Foundation created and the coalitions in
the US and UK to ensure all children have equal access to protection against these Can￿rs. The US
achieved the milestone of re￿MMendIng HPV protscb.on to all fjhiklren in 2011. Following the Foundation's
five-year advocacy campaign in the UK, sch(K>l-based HPV vaccination for began in September 2019.
Our focus now turns lo securing the same protection for W as well as gir15 across Europe.
While we work lo prevent future cases. it is also fundamental ihat scienlific and medical disparities are
addressed for the FK)pulalion that will continue lo be diagnosed wth HPV-related Can￿rS like anal cancer.
Treatment optsons have barely progressed since the 1970s and minimal research has been undertaken in
new therapies. The medical and scientific communities [￿ed better research tools and investment in novel
ideas to develop a Cure.
The stigma surrounding anal cancer remains one of the ￿n1[al thallenges to improving outcomes for anal
cancer patients. Unfortunately, many medical centres prowde only limited information specrfic to the
experien￿ of anal cancer. Many patients f*1 ISO￿ted and lack the support and educational resources that
significantly improve quality of lrfe during and after treatment.

Gender-neulral HPV va￿l￿ation began in September 2019. h¢)wever no announcement %WdS made as to
whether a catch-up programme would be instigated for boys aged 1&18.
In its meeting in earfy Febnjary 2020, the JCVI decided not to recommend a c2tch-up vaccination programme
for older boys. HPV Action did as much as we could to advocate for a catch-up programme despite our
limited capacity including meeting wrth JCVIIPHE in January 2020. wrreswndence with JCVI, the
engagement of several MPS who wrote letters to minislers and tabled PQS, and a social media campaign
through twitter and FaceLK)ok. Whilst this was a disappointing outcome it was felt that it was not worth
Continuing with this effort because it was extremely unlikely that JCVI would change its mind, especially in the

In the UK, about 1,500 people, and in the US, about 8,600 are diagnosed wth anal cancer annually. Anal

one or muf(iple errant ￿lIS to Multip￿ until they affecl the functioning of the body. Immunotherapies seek to
address this rnalfuncb.on by m¢xlulab"ng spectfic aspects of Ihe immune syslem to tsrget can￿r cells.
Supporting RèsÈareh to Find A Curè for multip￿ HPV*ÈlatÈd Cancè
HPv-re￿led cancers represent SW. of the world's cancers and treatment to cure these rllalignancies has
barely advanced in decadès. To add￿sS this vast and unmet need, the Foundation partnered VAth Stand Up
To Cancer and the Farrah Fawcett Foundation in a $1.2 million grant to support researchers at Dana-Farber
Cancer Institute in Boslon, Ma55achusetts, vtho are irwesb"galing the treatment of HPV-relaled canGer by
harnessing the immune system against it. The study was a first of its kind as we sought a therapy to help
multiple HPV-related cancers, instead of addressing each cancer individualty.
The sludy focuses on the preclinical and clinical development ol an immunotherapy Ireatment for patients
wrth recurrent HPV-related anal. cervical, and head and neck cancers who have relapsed after their initial
therapy as such patients have few therapeutic oplions available lo them. A successfLJI vaccine would be used
to augment chemotherapeutic outcomes and improve Surwval. Specifically, this Phase 1 b12 trial tests tumor
response and safetylefficacy of vaccination with E711-19 nanomer for the t￿atment of incurable HPV-16
related head & neck, cervical, and anal can￿r in HLA-A2 ￿sth'Ve patienis. It investigates the theory of
therapeutic CD8 vaccines against HPV epf(opes. This project is W by Ellis L. Reinherz, M.D., Chief of the
Laboratory of Immunobiology and Co-Dire¢tor of the Cancer Va¢¢ine Center at Dana-Farber and Robert l.
Haddad, M.D., Disease Center Leader for Head and Neck Ooncology at Dana-Farber.

OBJECTIVES AND ACTMTIES
The trial was completed in 2019 and resulted in three publications. Whilst the trial agent produced no durable
responsè in study participants, rèsearchers StUd￿ng the resuts wèie ablé to gain a better understanding of
the diversity of cancer cells in HPV cancers as well as why certain HPV therapeutic vaccines might fail.
Additionally, the tri81 improved care in the HPV can￿r space by creating connections among HPV laboratory
and clinical investigators across the Boston research community, and more broadly, across the national HPV
advocacy communty. As the first trial lo investigate a IherapEulic vaccine for mU￿1Ple HPV cancers. it was a
critical first step towards unif￿ng a once-siloed research community to find a cure for all HPV cancers. As a
result, where once each HPV cancer was researched solely in individual trials. Ihere are now multiple clinical
trials specifically researching agents to treat and cure all HPV Can￿rs.
Encouraging the scientific field to view these cancers comprehensNely resutted in a new approach for
gèn8rating promising treatments on behalf of the hundreds of thousands of people living with HPV-rèlatéd
cancers. In light of this significant shift led by the Foundation, there are now several phase l and 11 trials
underway that seek to find a treatment for Multip￿ HPV-related cancers al the same lime.
LOOKING AHEAD: 2021
As we Iwk ahead. the HPV and Anal Cancer Foundati(￿ Continues to seek the most effective methods to
prevent HPV and cure the cancers it causes.
The Foundation wll continue to work tirelessly to eradicate all HPV-related cancers by furthering public wlicy
and scientific debate, eliciting further professional and expert supwrt, encouraging the development of
regional initiatives on vaccinating bo￿. and demonstrating why it is important to protect all children against
HPV at the recommended age.
Although the UK b￿an vaccinating boys against HPV as pupils retum to schools in September 2019, we will
continue to work with Public Heatth England and other Stake￿)lderS in the HPV-related disease space to
ensure that uptake for this cancer-prevenb.ng intervention among 12-13 year old boys is as high as possible.
The interruplion caused by the pandemic wll undoubledly affecl vaccination rates in the UK and beyond. In
the wake of a growng vaccine hesitancy senlimenl. our work is more important than ever.
Our advocacy focus now turns trjwards tMJikJing our nascent partnership with the European Cancer
Organisabon and the HPV Action Network in order to achieve change across the European wntinent. The EU
Beating Cancer Plan set to be announced in 2021 ￿11 a￿d￿sS cancer-related inequalities between and within
Member States with actions to supw)rt, coordinate. and complement Member States, efforts. With over 40 /.
of ¢an¢ers in the region preventsble, the extension of HPV va¢¢inatKJn programmes to prote¢t boys as well as
girls provides a recognised and effective means to tackle cancer.
Through the NOMAN is an Island Campaign, collatrx)rations wth adV(￿teS and legislators, media attention
and athleb'c event participation, a broader audience is understanding the importance of protecting all children
against HPV_ With the NOMAN side of the organisation leading on HPV-related adivities. our platform is
growing from which to broadcast our message. In 2021, we wll continue lo partner with ihe Haute Route on
further cycling events as their European Charity Pather. furthering the reach of our message to prevent 5 /.
of cancer, and providing the opportunity to reconnect with the cyclists wthin our community. Additionalty, the
return of our flagship Bar￿Iona - Ibiza.. Row to End HPV races will as ever, capture the imagination of, and
spread the message ol how we can ￿￿vent HPV ¢an¢ers, to a broader audience.
We remain committed to finding better cures for patients and creating the essential research building blocks
that currently do not exist. For four de(2des. tréatment for all stages of anal cAncer had remained essentially
Ihe same with no FDA-approved drugs targeted and the same toxic side effects as in the 1970's. Our
breakthrough with MD Anderson Cancer Center is offering new hope to anal Gancer p8lien15 and we look
forward to ftjrther progress in the search for a cure.
