Oeteclion, involves working lo idenlify glycan biomarkers that will offer novel largtsls for therapy. In parallel, we aro

arriv￿ al Ih8S8 siles. 11 h8s beer) hypnlhesised that lumours help prepare these locations lo make them morel
susceptible lo future tumour spre8d. In particular. it was observed that immune cells, especially neutrophils,
incr8as8 in these "pre-melastalic oryans, showing early pr8paralion for cancer spr8ad. end a posslbl8 role for
nBulrophils in thi8 process.
Enhancing immunotherapy wlth metfomiin
Further progres5 was made wSlh data $howing that the diabetes drng matformin improves the effectiveness of an￿-
canc8r immune Iheraples, particularly in mi¢9 with diel-induced [￿9$11y. By studying lurnours from these mice, th&
team found that melformln-tre8ted lumours in obese mlce had more cancer-fighling immune cells, called CD8 T-
cells, which may h8lp Slow tumour growth. This is being pgrformed In collaboration with Dr Simon Lord. ABC Senior
Research Fellow, Orièl College, Unlver8ily of Oxford.

Models of obesity for tha study of br8891 caiiLer wwe further charac18rised lo 8ngure that they 8re physiologically
rel&vant and comparable lo human cllnical data. Advanced imaging lo measure 181 distribution in obese micg,

Southampton 188m produced a special form of a canc8r largeling antibody using this new method, shDwing il works

New Grant Incorne and Investment
Max Crispin received new funding frorn the Bill & Melind8 Gates Foundation of £1 .4M over 2.5 year5, a 9ranl that
will help support the Gore infrastructure of the group removing potential pressures on the ABC programme. Max
and his Post4Joc Jo85 w8re also awarded $200,000 over 2 years from the Inslilule of pro18in design, funded by the
Gales Foundation, lo further support the general running of the lab and r859arch environment.

cells. Dr Nicho185 is using sophisticated techniques lo develop neoantigen vaccines, by training T-cells lo identify
lumor-specific mulalions and mount a largeled immune response. specific lo cancer cells. A5 part of a CRUK
network accelerator, the leam developed a proleogenomic slralegy lo improve neoanligen identific81ion and
achieved Immunogenic responses in 5 of 8 patients (80Yo).
Ongoing work 15 focusing on the potential effect of phagocytosis ('cell eating,) and consequent high inlracellular
lipid on IriK￿e negativ8 breast cancer efferocylosis (dying cell removal) and peptide presenialion lo T-cells by MHC
molecules. This is important because TNBC presents unlque challenges for direct idenlificalion of T-cell

back office procedures. Thls was a huge slep lorw8rd lor the charity, saving a consid8rable amount of time for all
concerned.
In addition to linking to the charity database, the website can also now directly process paymants wilhoul th8

FINANCIAL REVIEW
During th8 financial year ended 2024 Against Breast Cancer continued lo remain committed to achieving its
objeclives in support of financial growth and stability despite a challenging economic environment. As a result, a
profil of £104,179 was made in 2024 compared to a loss of1£63,2081 In 2023. Throughout the year a continued
uplifi could be 58en in fundraising with events being at it5 stronge51 post pandernlc and legacles showing an upward
movem8nt in revenue performanc8. This is reflecl8d in th8 con501idat8d income for 2024 being £1,024,382 in
contrast to 1,026,990 in 2023. The charily15 moving even closer to meeting ils income growth strategy of increasing
inGome to £1.5 million in the next four yearg, so enabllng further Spend on research.

Following the funded research of tha Dielcomplyf study al the University of Weslminsler in 2015 the chafity
developed a r8se8rch strategy funding lono term and short-term innovative rèsearch projects. In addition to our
long term core research we also fund PhD's and Junior Research F8llowghips for the advancement of education
through teaching and ragearch. Thes6 projects are I￿r￿ed responsibly in line with Against Breast Cancar objectives,
and surplus funds ring fenced annually for further spend on research proj8cts and lab equipment. The res&arch
strategy and charity long lemi projections see responsible, sustainabl8 and substantS8l growth in funding in line with
projected growth In Income and maximising ROI.

in 2018 Against Brea81 Cancer with Oriel College sel up a 9 year ABC funded Junior Fellowship programme to

employees, professlonal fees and a wind down of the researGh programrne.
