At Oriel College in Oxford, Senior Research Fellow Dr Sirnon Lord was gwen the honour of being guest S￿aker at
the colleg8's Alumai We8kend. Already havirvJ rea¢h8d a prominent role in dinicd brea￿ eancar resèarch
in Oxford, Dr Lord is expected to become a leader in clinical breast cancer research.

A new study from the Un￿ersty of Southampton has discovered that 'crown4ike structures, surrounrfing breast
tumours in oven¥eighi and obese patients could hinder their response to therapy.

overexpre5sed breast cancer.
Adipose tissuè, or b(￿Y fat, is an irnportant comwnent of the hèalthy human breast and y8f high thdy mass index
IBMII is assouated with increased risk of developing brea* cancer. Overwaght patients aL80 have worse survNal
rates than patients wlth h&aW body weight.
In patients with a high BMI, irEcreased body fat surrounding the breastcan cause inflammatory irnmune cdls, called
rnacTophages, to gather in the breast's fat tissue. Trese macrophages ¢an then forrn what are called 'wwn-like
structures. by surrounding thes8 fat cells (see picture below). This Creates an inflatnmatory environment in the
breast which can lead io the onset and growth of turnours.
How these crown-1Skè struclures go on to affect breast canc8rprogressk)n and response to therapywas prevSous
largely unknown.
The research team, led by Professors Stephen Beers, Ramsey Cutr8S5 and Aganst Breast Cancerfunded Lecturer
Dr Chades Birts. assessed samples from a gr￿p of HER2+ breast earKer patients to investigate the link b8tween
high BMI and Ihe forrnation of crown-like structures, a￿d the subsequ8nt eff8cl ofthese on how patients ￿spOndE￿
to therapy wth a dnjg caled trastuzurnab IHerceptirl&l.
The results, published in the Journ￿ Scientific Reports, showed Ihat patients that were overweight or obese had
signilicanlly MO￿ CTown-like structur8S in their lat tissue surrounding the turnour, and thatthis was assouated with
a faster time to metastalc disease, an indleation of how well the pat'ents have responded to therapy.
They then went on to klentify a PDtential molecular biomarker, cJled CD32B. on the surtace of the macrophages
in these crown-like structures. Wh8n this marf(er was present in ovèrweight and obese patients, their response to
trastuzurnab therapywas [￿rer.
Professor Beers explai￿, Yhese ffndings wll be of interest to dinioan5 and researchers invofved in breast cancer
tr8atment as they coukl potentially be used to develop p8rsonallsed treatrnent in Pthnts with HER2+
overèxpressed breast caNcer.'
"For eXam￿e, doctors woukj know that patients with a high BMI and CD32B on theircrown4ike structu￿$ are I1￿elY
to have a poor r8sponse totrasbJzumabtherBpy. They rnaytherefo￿ benefitfrorn more intensive8nti-HER2therapy
earfier in thwr treatment.
'On Ihe other hand, this Study highligms how effective trasiuzumab treatment is in patients that do not have CD32B
Posthve crown-like structures. So these patients coukl benefit froln a lower dose of &nti-HER2 therapy which may
minimise the sidè-effects the experience. Further studies wth MO￿ patients wll be needed to hdp confirm these
initial firKlings.'
The research team are now looking at ways to change the bèhamour of these CTown-like stnjctures to improve

awareness and advice. we added new lools and created new resourc8$ to enable us to better meet needs of

We continue to grow our digital presence and plan to continue on this trajectory. Our brand is TncTÈasingly

In 2022 we had the unique opportunity to ￿eruit two London Marathon tearns for events in OciobEr aThl April 2023.

Our Play it Pink campaign proved popular 5POrts dubs. team England hockey ￿aYer Sarn Ward jointy raised
over£701K> together wrth his local tsam in Bishops Stortford.
Locally. our first In-[￿rsOn event was to fu￿1 a commibnent made in 2019 to the Harwell Women's Institute. The
group has been supporting the charity since the beginning. Annual events Splash of Pink aNd Witney in the Pink
retumed, a￿ayS popular, they each provide great Opportun￿9$ to mwntain our protila in Oxfordshire and PTOVkYe
an important SOUT￿ ol funds.
We welcomed a new ind8peThJent fundrnsing group Charity AngeLs, pledg6d to fund Sirnon Lord's Tesearth project

Cancer Awareness Month. This support takes many lorms. from pink products and packaging to In-house

FINANCIAL REVIEW
Against Breast Cancer made a profit of1£17,5521 in 2022 comparwj to a loss 011£156,9181 in 2021. Consolidated
inwme for 2022 was £875,116 compared to £754,767 in 2021. Income was for a third year dramatirally impacted
but showed a marked increase in funds indicating that we are entering into a rnore stable peric¥J. As w8 move into
2023 there i8 a delinrfte uplift in fundraigng wih evèrrts now back to amost a pre pandern￿ level. Edging closerto
bringing us back in line ￿th the 5-year income grobvth strategy of 2019 with the intention of in¢￿asIng incDme to
£1.5 million overthe next ￿uPle of years.
Followng the funded research ol the DietCom￿y[ st￿Y at the Unwer5ity of Westminster in 2015 the chatity
developed a research strategy funding long term and short-ierrn innovatwe research prolects. In addition to our
loNJ terrn cor8 research we also fund PhD's and Junior Research Fellowships tor the advancernent ot education
through teaching and research. These projects are funded responsibly in line with Agwnst Breast Cancers
objectives, and surplus funds ring fenced annualy for further spend on researth projects and lab equipment. The

Against Breast Cancer continues to work ￿th the clinical 4)onsor ot thè D]'etCompLyf sludy, University College

search. Against Breast Can￿r ha5 a projected long-term res8arch portfolio in excèss of £4million. This portfolio
has been steadily built in line wiEh the ie58arch strategy arKI eharity long term projections 8nsuriry sustainabiltty for
the future.
PLANS FOR FUTURE PERIODS
Looking forward to 2023 we have already recruitsd the largest nurnber of overseas events partiupants the eharEty

Cancer achiev88 its objectNe of commissionlng research through rts close relationship with a research team based
at Unpiersty of Southampton, and a research team based at the Universty ol Oxford. The charity promdes a grant
to each t8arn which funds ihelr salary and laboratory and enables th8 or¥Joing ￿SearCh to take pla￿.
Ag&llst Breast Cancer also established a Fellowship programme in 2014, to support rising scientific talent in the
b￿ast cancer research Ileld. The first 3-year Fellowship i8 associated wth Oriel College, Oxford. and was award£d
to Dr. Max Crispin who is designing newtreatrxents for metastabc b￿ast cancer. Following on from this Fellowship.
in 2018 Aganst Breast Cancer wth Oriel College s&t up a 9 year ABC funded Junior Fellowship programme to
continue advancements of education through t8aching and research.
